Description
Tryptose Sulfite Cycloserine Agar Base (TSC) – For Effective Detection of Clostridium perfringens
Product Overview
Tryptose Sulfite Cycloserine Agar Base (TSC) is a vital culture medium specifically designed for the detection and enumeration of Clostridium perfringens in food products. Moreover, this medium is particularly effective when combined with 50% Egg Yolk Emulsion, which provides an optimal environment for isolating this important pathogen.
Key Ingredients
Ingredient | Amount (gm/liter) |
---|---|
Pancreatic Digest of Casein | 15.0 |
Soy Peptone | 5.0 |
Yeast Extract | 5.0 |
Sodium Metabisulfite | 1.0 |
Ferric Ammonium Citrate | 1.0 |
Agar | 20.0 |
Final pH: 7.6 ± 0.2 at 25°C
Preparation Instructions
- Suspension: First, suspend 47.0 g of TSC in 1 liter of distilled or deionized water.
- Mixing: Next, mix thoroughly and heat with frequent agitation, boiling for 1 minute to ensure complete dissolution.
- Autoclaving: After that, autoclave at 121°C for 15 minutes.
- Cooling: Once cooled to 50°C, add 20 mL of D-cycloserine to the medium and mix well.
- Egg Yolk Plates: For egg yolk plates, additionally add 20 mL of 50% Egg Yolk Emulsion to 250 mL of the prepared medium.
Principle
TSC provides essential nutrients, including carbon, nitrogen, vitamins, and minerals, that promote the growth of Clostridium perfringens. Furthermore, the yeast extract supplies B-complex vitamins, thereby enhancing bacterial growth. Sodium metabisulfite and ferric ammonium citrate act as indicators for hydrogen sulfide production, forming a characteristic black precipitate upon sulfide reaction.
Appearance
- Dehydrated Medium: The dehydrated medium appears as a free-flowing yellowish powder.
- Prepared Medium: In contrast, the prepared medium manifests as a yellowish solid.
Storage Conditions
- Dehydrated Medium: Store tightly capped at 5-30°C. In addition, the shelf life is 3 years from the manufacturing date.
- Prepared Medium: Conversely, store at 2-8°C, away from direct light.
Quality Control
To ensure quality control, incubate inoculated plates at 36±1°C for 20-24 hours to confirm that the medium effectively supports the growth of Clostridium perfringens.
Conclusion
In summary, Tryptose Sulfite Cycloserine Agar Base (TSC) is essential for laboratories engaged in food safety testing and microbiological research. Ultimately, its specialized formulation allows for the reliable detection of Clostridium perfringens, thus helping to ensure food quality and safety.
