Description
Mueller-Hinton Broth (MHB) is a premium nutrient-rich medium widely used in microbiology laboratories for antimicrobial susceptibility testing and the cultivation of various microorganisms. With a balanced formulation, MHB supports the accurate evaluation of antibiotic efficacy, making it an essential tool for researchers, clinical labs, and educational institutions.
Key Features:
- Nutrient Composition: Contains:
- Beef Extract (3.0 g/L): Provides essential nitrogen and carbon sources.
- Soluble Starch (1.5 g/L): Acts as a protective colloid against toxic substances and serves as an energy source through hydrolysis.
- Acid Hydrolyzate of Casein (17.5 g/L): Supplies vital amino acids, nitrogen, and sulfur.
- pH Level: Final pH of 7.4 ± 0.2 at 25°C ensures optimal growth conditions for a wide range of bacteria.
- Low Thymine and Thymidine Content: Ideal for broth dilution antimicrobial susceptibility testing, allowing for precise assessment of antimicrobial agents.
Preparation Instructions:
- Suspend 22.0 g of Mueller-Hinton Broth in 1 liter of distilled or deionized water.
- Mix thoroughly and heat with frequent agitation until dissolved.
- Sterilize in an autoclave at 121°C (15 lbs) for 15 minutes.
Appearance:
- Dehydrated Medium: Free-flowing yellowish powder.
- Prepared Medium: Yellowish transparent liquid.
Storage Conditions:
- Shelf Life: Dehydrated medium: 3 years from the date of manufacture. Prepared medium: Store at 2-8°C away from direct light.
- Storage Instructions: Keep tightly capped in the original container at 5-30°C. Do not use if past shelf life, caking, or color variation occurs.
Quality Control:
Cultural characteristics should be assessed after incubation at 35-37°C for 18-24 hours to ensure proper functionality and performance.
Applications:
- Antimicrobial Susceptibility Testing: Widely recognized for use in determining the effectiveness of antibiotics against bacterial pathogens.
- Microbial Cultivation: Suitable for the general cultivation of a variety of bacteria in laboratory settings.
