Description
N6 Medium is a specialized formulation designed for optimal plant tissue culture. This versatile medium supports the growth of various plant species, making it an essential tool for researchers and horticulturists alike. With a composition that excludes agar, N6 Medium provides a unique solution for in vitro cultivation.
Key Features and Composition
N6 Medium consists of a balanced mixture of essential nutrients that promote healthy plant growth. Each liter of the medium contains:
- Potassium Nitrate: 2800 mg
- Ammonium Sulfate: 463 mg
- Dipotassium Phosphate: 400 mg
- Magnesium Sulfate (MgSO4·7H2O): 185 mg
- Calcium Chloride (CaCl2·2H2O): 165 mg
- EDTA Disodium Salt: 37.3 mg
- Ferrous Sulfate (FeSO4·7H2O): 27.8 mg
- Manganese Sulfate (MnSO4·H2O): 4.4 mg
- Zinc Sulfate (ZnSO4·7H2O): 1.5 mg
- Boric Acid: 1.6 mg
- Potassium Iodide: 0.8 mg
- Vitamin B1 (Thiamine Hydrochloride): 1.0 mg
- Vitamin B6 (Pyridoxine Hydrochloride): 0.5 mg
- Niacin: 0.5 mg
- Glycine: 2.0 mg
- Sucrose: 20,000 mg
The pH of N6 Medium is optimized at 5.8, ensuring ideal conditions for tissue culture.
Usage Instructions
To prepare N6 Medium, dissolve 24.1 g of the powder in 1000 ml of distilled water. Heat and stir until completely dissolved. Then, dispense the solution into flasks and autoclave at 115°C for 20 minutes. If the pH is too low, adjust it using diluted sodium hydroxide to reach the desired level of 5.8.
Applications and Benefits
N6 Medium is widely used in plant tissue culture laboratories due to its efficacy in supporting cell division and growth. It is particularly beneficial for studies involving plant regeneration, micropropagation, and genetic transformation. By providing essential macro and micronutrients, this medium enhances the growth potential of various plant species.
Researchers appreciate the all-inorganic salt formulation, which minimizes contamination risks. Additionally, a slight precipitate may form post-sterilization, which does not impact the medium’s effectiveness.
Microbial Sensitivity Testing
N6 Medium is also suitable for microbial sensitivity testing. By inoculating with quality control strains at 10-100 CFU, users can assess the medium’s ability to support growth while maintaining sterility. visit our Plant Tissue Medium Series for more related products
