Directions for Use
1. Suspend 27.0 g of the base medium powder in 1000 mL of distilled water.
2. Add an appropriate amount of liver pieces (typically 5-10% v/v) to the solution.
3. Heat to boiling to completely dissolve the powder.
4. Sterilize by autoclaving at 116°C for 40 minutes.
5. Cool to room temperature under anaerobic conditions before use.
Microbial Sensitivity Test
Test Protocol: Inoculate the QC strain and incubate at 36±1°C for 18–24 hours under anaerobic conditions.
QC Strain: Clostridium chauvoei (Gas Gangrene Bacillus)
9-Tube Method: Control/Sample: Growth observed in all 3 replicates at dilutions 10⁻⁶, 10⁻⁷, and 10⁻⁸.
Plate Count Method: Control: TNTC at 10⁻⁵ & 10⁻⁶; 63/66 at 10⁻⁷. Sample: TNTC at 10⁻⁵ & 10⁻⁶; 58/64 at 10⁻⁷.
