Description

Staphylococcus Selective Agar: Your Essential Medium for Isolating Staphylococci

Staphylococcus Selective Agar is a premier medium specifically formulated for the isolation and differentiation of staphylococci. This essential tool in microbiological laboratories leverages mannitol fermentation and pigment formation to provide accurate identification of bacterial strains.

Key Features

  • Selective Growth Conditions: The high sodium chloride concentration, combined with a specialized selective supplement, effectively inhibits most non-staphylococcal bacteria. This creates an optimal environment for staphylococci to thrive, ensuring precise results.

  • Mannitol Fermentation: D-mannitol serves as a critical carbohydrate source, allowing pathogenic coagulase-positive staphylococci to ferment it and produce acid. This fermentation leads to noticeable color changes in the agar, aiding quick differentiation.

  • Distinct Pigment Formation: Pathogenic staphylococci typically yield yellow-orange colonies, providing a visual cue that simplifies identification and enhances the reliability of results.

Composition and Preparation

Each liter of Staphylococcus Selective Agar comprises:

  • Pancreatic Digests of Casein (10g): A rich source of nitrogen, vitamins, and minerals that promotes robust bacterial growth.
  • Peptic Digest of Animal Tissue (10g): Further supports microbial proliferation.
  • Beef Extract (2g): Supplies additional vitamins necessary for optimal growth.
  • D-Mannitol (10g): Facilitates fermentation tests critical for identifying pathogenic strains.
  • Sodium Chloride (40g): Establishes the selective environment crucial for isolating staphylococci.
  • Bromocresol Purple (0.02g): Serves as a reliable pH indicator.

To prepare the medium, suspend 102g in one liter of distilled water, heat to dissolve thoroughly, and autoclave at 121°C for 15 minutes. For best results, store the prepared medium at 2-8°C, away from direct light.

Quality Control and Storage

For effective quality control, incubate inoculated samples at 36±1°C for 24-48 hours. Staphylococcus Selective Agar has a shelf life of three years when stored properly at 5-30°C. Do not use any caked or discolored medium, as this may compromise experimental integrity.
