- Pancreatic Digest of Casein (5.0 g/l): Provides vital amino acids and nitrogenous compounds essential for robust bacterial growth.
- Yeast Extract (2.5 g/l): Enriches the medium with Vitamin B complex, promoting the growth of diverse microbial populations.
- Dextrose (1.0 g/l): Supplies necessary sugars for enhanced microbial proliferation.
- Agar (15.0 g/l): Acts as a solidifying agent, forming a stable gel for reliable colony counting.
Preparation Instructions:
To prepare the medium, suspend 23.5 g of Plate Count Agar in 1 liter of deionized or distilled water. Heat with frequent agitation until fully dissolved, then boil for 1 minute. Dispense into sterile tubes or flasks and sterilize in an autoclave at 121°C (15 lbs.) for 15 minutes.

Plate Count Agar serves as the go-to medium for the plate count of microorganisms in food and other materials. Following the APHA pour plate method, diluted samples are added to Petri plates before sterile molten agar is introduced, ensuring even mixing. This method provides higher recovery rates compared to surface inoculation, making it ideal for applications in sterile environments and laboratories.

Storage and Shelf Life:
Store tightly capped in the original container at 5-30°C. The dehydrated medium boasts a shelf life of 3 years from the manufacturing date, while the prepared medium can be stored away from light at 2-8°C.
Quality Control:
Cultural characteristics should be evaluated after incubation at 35-37°C for 18-48 hours, ensuring optimal performance for microbial analysis.
