Lactose Broth for Coliform Detection and Lactose Fermentation

Lactose Broth is a versatile culture medium that plays a crucial role in microbiological analysis. It is specifically designed for the detection of coliform organisms, serving as a pre-enrichment broth for salmonellae, and facilitating the study of lactose fermentation across various bacterial species. This high-quality medium provides essential nutrients, ensuring robust growth and accurate results in laboratory settings.

Ingredients and Preparation

The formula includes:

  • Pancreatic Digest of Gelatin (5.0 g/liter): Supplies vital nutrients for bacterial metabolism.
  • Lactose (5.0 g/liter): Acts as a fermentable carbohydrate, particularly beneficial for coliform bacteria.
  • Beef Extract (3.0 g/liter): Enhances the growth environment with additional nutrients.

To prepare, simply suspend 13.0 g of the powder in 1 liter of purified water. Warm gently until completely dissolved, then dispense into test tubes containing inverted Durham tubes. Sterilize using an autoclave at 121°C for 15 minutes. Afterward, cool the broth quickly to maintain its integrity.

Principle and Interpretation

Lactose Broth’s unique composition allows for effective testing of coliform presence. When bacteria ferment lactose, they produce gas, indicating a positive reaction. This process makes it an ideal choice for conducting presumptive tests for coliforms, ensuring reliable and accurate results in your microbiological studies.

Appearance and Storage

This dehydrated medium appears as a free-flowing yellowish powder, while the prepared medium becomes a yellowish transparent liquid. For optimal shelf life, store Lactose Broth tightly capped in its original container at temperatures between 5-30°C. The dehydrated form remains usable for three years from the manufacturing date, while prepared medium should be kept at 2-8°C, protected from direct light.

Quality Control

To ensure effectiveness, inoculate and incubate the prepared broth at 36±1°C for 18-24 hours. After incubation, add 1-2 drops of 1% phenol red solution to observe acid production, confirming the presence of coliform organisms.
