- 5-bromo-4-chloro-3-indolyl-α-D-glucopyranoside: 0.1
- Agar: 15.0
Final pH: 7.0-7.2 at 25°C
Directions for Preparation
- Suspend 8.62 g of the medium in 200 mL of distilled water.
- Sterilize by autoclaving at 121°C for 15 minutes.
- Cool to approximately 50°C and pour into sterile Petri dishes.
Procedures for Use
- Inoculate 10 μL of MSLT-Vm broth or EE enrichment broth onto the chromogenic plates using a sterile inoculation loop.
- Invert the plates and incubate at 36 ± 1°C for 22-26 hours.
- Examine the plates; typical Enterobacter colonies will appear as blue-green.
Quality Control
- Appearance:
- Dehydrated powder: homogeneous, with good flowability, yellowish or milky white.
- Prepared medium: yellowish solid.
- Microbial Sensitivity Test:
- Incubation at 36 ± 1°C for 18-24 hours should yield blue-green colonies of sakazakii and colorless or yellow colonies for other bacteria.
Storage Recommendations
- Prepared Plates: Keep for 2-5 days; avoid direct light exposure.
- Powder Medium: Store in a dark, dry place at 2-8°C; protect from light.
Failure Criteria
Do not use dry powder media that are past their shelf life, exhibit caking, or show color variations.
