Description
DNase Agar – Essential Differential Medium for Bacterial Identification
Product Overview
Discover the power of DNase Agar, a premier differential medium expertly formulated for the detection of deoxyribonuclease activity in bacteria. Perfect for clinical diagnostics and microbiology research, this medium facilitates the accurate identification of pathogenic bacteria from clinical specimens.
Key Features
- Rapid Detection: Easily identify DNase-positive bacteria by observing clear zones after treatment with 1N HCl.
- Enhanced Visualization: Utilize 0.1% toluidine blue solution for even clearer results; positive DNase activity appears as pink to red zones, while negative results remain blue.
- High-Quality Ingredients: Made with pancreatic digest of casein and peptic digest of soybean meal to ensure optimal growth conditions.
- Long Shelf Life: Store at 5-30°C with a shelf life of 3 years, ensuring reliable performance in your lab.
Ingredients and Composition
Ingredient | gm/liter |
---|---|
Pancreatic Digest of Casein | 15.0 |
Peptic Digest of Soybean Meal | 5.0 |
Calcium Chloride | 0.02 |
Sodium Chloride | 5.0 |
DNA | 2.0 |
Agar | 12.0 |
Final pH | 7.3±0.2 |
Preparation Instructions
- Suspend 42.0g of DNase Agar in 1 liter of distilled or deionized water.
- Mix well and heat with frequent agitation until fully dissolved.
- Dispense into triangle flasks and sterilize in an autoclave at 116°C for 15 minutes.
- Cool to 45°C and pour into sterile petri plates.
Testing Protocol
- After incubation at 37±1°C for 18-24 hours, flood the plates with 1N HCl and observe for clear zones indicating DNase activity.
- For DNase Test Agar with Methyl Green, reagent addition is not required.
Principle and Interpretation
The pancreatic digest of casein and peptic digest of soybean meal provide necessary nutrients for bacterial growth, while DNA serves as the substrate for DNase activity. The presence of clear zones around bacterial colonies after flooding with HCl indicates DNase activity, confirming the identification of DNase-positive bacteria.
Appearance
- Dehydrated Medium: Free-flowing yellowish powder.
- Prepared Medium: Yellowish gel.
Storage Instructions
- Store tightly capped at 5-30°C.
- Dehydrated medium has a 3-year shelf life.
- Prepared medium can be stored out of direct light at 2-8°C.
Quality Control
Ensure accurate results by inoculating and incubating at 37±1°C for 18-24 hours. Flood with 0.1% Toluidine Blue solution after incubation to visualize DNase activity.
