Cetrimide Agar is a highly effective growth medium specifically formulated for the selective isolation and presumptive identification of Pseudomonas aeruginosa, a critical pathogen in both clinical and environmental microbiology. This medium is enriched with essential nutrients to support the growth of this organism while inhibiting competing bacteria.
Key Ingredients:
- Pancreatic Digest of Gelatin (20.0 g/L): Provides essential nutrients for robust growth.
- Magnesium Chloride (1.4 g/L): Stimulates pyocyanin production, a vital identifier for Pseudomonas aeruginosa.
- Potassium Sulfate (10.0 g/L): Enhances the growth and metabolic activity of the target organism.
- Cetrimide (0.3 g/L): A quaternary ammonium compound that selectively inhibits non-target bacterial species.
- Agar (13.6 g/L): Acts as a solidifying agent for the medium.
Final pH: 7.2±0.2 at 25℃
Preparation Instructions
To prepare, suspend 45.3 g of the dehydrated medium in 1 liter of distilled or deionized water, add 10 ml of glycerol, mix thoroughly, and heat with frequent agitation. Dispense into flasks and sterilize in an autoclave at 121℃ for 15 minutes.
Appearance
- Dehydrated Medium: Free-flowing yellowish powder.
- Prepared Medium: Milky-white gel.
Storage and Shelf Life
Store the medium tightly capped in its original container at 5-30°C. The dehydrated form has a shelf life of 3 years from the date of manufacture. Prepared medium should be stored out of direct light at 2-8°C.
Quality Control
Inoculate and incubate at 30-35℃ for 24-48 hours to ensure optimal growth conditions.
