Description

Hugh Leifson Agar for Aerobic and Anaerobic Bacterial Testing | Essential Microbiology Medium for Glucose Metabolism

Detailed Product Description:

Unlock the secrets of bacterial metabolism with Hugh Leifson Agar, a specialized medium designed for microbiologists focused on differentiating aerobic and anaerobic carbohydrate breakdown. This versatile agar is perfect for studying gram-negative intestinal bacteria, making it an essential tool in both clinical and research laboratories.

Key Features:

  • High Carbohydrate Concentration: Facilitates accurate differentiation of fermentative and oxidative metabolic processes, ideal for identifying bacteria in clinical samples.
  • pH Control: Incorporates dipotassium phosphate to maintain stable pH levels, ensuring optimal conditions for microbial growth and activity.
  • Motility Assessment: The agar consistency allows for easy observation of bacterial motility, providing a comprehensive analysis of microbial behavior.
  • Color Change Indicator: Clear color shift from greenish-blue to yellow indicates acid production, simplifying result interpretation.

Ingredients:

Ingredient gm/liter
Pancreatic digest of casein 2.0
Sodium chloride 5.0
Dipotassium phosphate 0.3
Bromothymol blue 0.08
Glucose 10
Agar 2.0
Final pH 6.8 ± 0.2 at 25°C

Preparation Instructions:

  1. Suspend 19.38g of Hugh Leifson Agar in 1 liter of distilled or deionized water.
  2. Mix well and heat with frequent agitation.
  3. Dispense into tubes and sterilize in an autoclave at 121°C (15 lbs.) for 15 minutes.
  4. Cool the medium in an upright position.

Principle and Interpretation:

Developed by Hugh and Leifson, this agar medium utilizes the taxonomic significance of carbohydrate metabolism. Microorganisms can utilize carbohydrates through fermentation or oxidation, and this property is crucial for bacterial differentiation.

  • Fermentative Organisms: Produce acid in both sealed and unsealed tubes, indicating metabolic activity.
  • Oxidative Organisms: Generate acid in unsealed tubes with minimal growth in sealed tubes.

Appearance:

  • Dehydrated Medium: A free-flowing yellowish powder.
  • Prepared Medium: A yellowish transparent gel.

Storage Conditions:

  • Store tightly capped in the original container at 5-30°C.
  • Dehydrated medium shelf life: 3 years from the manufacturing date.
  • Prepared medium should be kept out of direct light at 2-8°C.

Quality Control:

Follow label directions for preparation and inoculate, incubating at 36±1°C for 18-48 hours to ensure the medium’s efficacy.
