Description

Urea Agar Base: Essential Medium for Urease Production Testing

Urea Agar Base is a critical culture medium used in microbiology for distinguishing organisms, particularly within the Enterobacteriaceae family, based on urease production. This medium is designed to facilitate accurate identification through observable color changes, making it an invaluable resource for laboratory professionals.

Key Features

  • Differentiation of Bacteria: Urea Agar Base effectively differentiates urease-producing bacteria. When organisms metabolize urea, they produce ammonia, which raises the pH and causes a color change in the medium.
  • Simple Preparation: To prepare Urea Agar Base, dissolve 21.0g of dehydrated powder in one liter of deionized or distilled water. Heat with agitation until boiling, then autoclave at 121°C for 15 minutes. This straightforward process ensures quick and reliable results.
  • Visual Indicator: The medium contains phenol red, which changes from yellow to red-pink in alkaline conditions, providing a clear visual indication of urease activity.

Composition

Urea Agar Base consists of several key ingredients:

  • Peptic Digest of Animal Tissue: 1.0 g
  • Sodium Chloride: 5.0 g
  • Dextrose: 1.0 g
  • Potassium Dihydrogen Phosphate: 2.0 g
  • Phenol Red: 0.012 g
  • Agar: 12.0 g

The final pH is adjusted to 6.8±0.2 at 25°C. The dehydrated medium appears as a light red powder, while the prepared medium is an orange-red transparent gel.

Storage and Shelf Life

Store Agar Base in a tightly capped container at a temperature between 5-30°C. The dehydrated medium has a shelf life of three years from the manufacturing date. Once prepared, keep the medium in the dark at 2-8°C for optimal performance.

Quality Control

For quality assurance, prepare the medium following the provided instructions, inoculate, and incubate at 36±1°C for 18-24 hours. Always inspect the medium for any signs of caking or discoloration before use to ensure accurate results.

Conclusion

In summary, UAB is essential for identifying urease-producing bacteria in laboratory settings. Its easy preparation, clear visual indicators, and effective differentiation capabilities make it a preferred choice for microbiologists. For reliable urease testing and comprehensive microbial analysis, Urea Agar Base is your best option.
