To prepare the medium, suspend 46.0 grams in 1 liter of distilled water. Heat the mixture to boiling. After cooling to about 45°C, add 20 ml of Iodine solution (I2 6g, KI 5g, distilled water 20 ml) and 10 ml of 0.1% Brilliant Green solution. This combination optimizes conditions for Salmonella growth.
The formulation includes pancreatic digest of casein, peptic digest of animal tissue, calcium carbonate, bile salts, and sodium thiosulfate. These ingredients enhance the medium’s effectiveness. Adjust the final pH to 8.4 ± 0.2 at 25°C to meet performance criteria.
For best results, supplement each bottle with three boxes of BABIO-4086a Iodine Solution and three boxes of BABIO-4086b 0.1% Brilliant Green Solution. This step ensures effective selective enrichment for Salmonella.
