Description
E. Coli Broth is a highly effective cultivation medium specifically designed for the detection and enumeration of coliforms, especially E. coli, in various samples including water and food. This medium is essential for laboratories conducting microbiology testing, water quality analysis, and food safety assessments.
Key Ingredients:
- Pancreatic Digest of Casein (20g/L): Provides vital nutrients for robust bacterial growth.
- Sodium Chloride (5g/L): Maintains osmotic balance.
- Lactose (5g/L): Serves as a fermentable carbohydrate for coliform fermentation.
- Dipotassium Hydrogen Phosphate (4g/L) & Potassium Dihydrogen Phosphate (1.5g/L): Buffers to stabilize pH at 6.9 ± 0.2.
- Bile Salt No.3 (1.5g/L): Inhibits gram-positive organisms to ensure specificity in detecting coliforms.
Usage Instructions:
- Suspend 37g of E. Coli Broth in 1L of deionized or distilled water and mix well.
- Heat the mixture until completely dissolved, then dispense into tubes with Durham gas collection tubes.
- Sterilize at 121°C for 15 minutes.
Interpretation:
E. Coli Broth allows for the detection of fecal coliforms and helps determine their source—whether fecal or non-fecal—when used as a confirmatory test. The medium supports the growth of coliform organisms at 37°C and isolates E. coli at higher temperatures (44.5°C or 45.5°C) for comprehensive analysis.
Appearance:
- Dehydrated Medium: Free-flowing yellowish powder.
- Prepared Medium: Yellowish transparent liquid.
Storage:
Store tightly capped at 5-30°C. Dehydrated medium has a shelf life of 3 years, while prepared medium can be kept at 2-8°C, away from direct light.
Quality Control:
Inoculate and incubate at 44.5 ± 0.2°C for 24-48 hours to verify performance.
