Overview
Christensen Urea Agar Base serves as a premier microbiological medium designed for differentiating urease-producing organisms, especially within the Enterobacteriaceae family. This agar medium facilitates precise urease detection, making it essential for microbiology laboratories.

Key Ingredients
Each liter of Christensen Urea Agar Base contains:

  • Peptic Digest of Animal Tissue: 1.0 g
  • Sodium Chloride: 5.0 g
  • Dextrose: 1.0 g
  • Potassium Dihydrogen Phosphate: 2.0 g
  • Phenol Red: 0.012 g
  • Agar: 15.0 g

The final pH is 6.8±0.2 at 25℃. To prepare the medium, suspend 24.0 g in one liter of deionized or distilled water. Heat while stirring, then sterilize in an autoclave at 121℃ for 15 minutes.

Principle of Urease Detection
This medium allows easy detection of urease production. When urease-positive organisms metabolize urea, they release ammonia. This reaction raises the pH and causes a color change to red-pink, thanks to the phenol red indicator. This transformation helps microbiologists differentiate between urease-producing and non-producing bacteria.

Storage and Quality Control
The dehydrated medium appears as a light red powder, while the prepared medium is an orange-red transparent gel. Store the tightly sealed container at 5-30°C for a shelf life of three years. After preparation, keep the medium at 2-8°C, away from direct light.
