Preparation Instructions

  1. Suspend 42.0g of DNase Agar in 1 liter of distilled or deionized water.
  2. Mix well and heat with frequent agitation until fully dissolved.
  3. Dispense into triangle flasks and sterilize in an autoclave at 116°C for 15 minutes.
  4. Cool to 45°C and pour into sterile petri plates.

Testing Protocol

  • After incubation at 37±1°C for 18-24 hours, flood the plates with 1N HCl and observe for clear zones indicating DNase activity.
  • For DNase Test Agar with Methyl Green, reagent addition is not required.

Principle and Interpretation

The pancreatic digest of casein and peptic digest of soybean meal provide necessary nutrients for bacterial growth, while DNA serves as the substrate for DNase activity. The presence of clear zones around bacterial colonies after flooding with HCl indicates DNase activity, confirming the identification of DNase-positive bacteria.
